Abstract The reliance on antibiotics in goat semen extenders to control microbial contamination has raised concerns about the emergence of antimicrobial resistance. This study aimed to evaluate the antibacterial efficacy of mulberry ( Morus alba ), roselle ( Hibiscus sabdariffa ), and aloe vera ( Aloe barbadensis ) extracts as natural alternatives to conventional antibiotics. Semen was collected from Boer crossbred bucks (n = 5), and bacterial isolates were subjected to susceptibility testing using the disc diffusion method. Semen samples were extended with plant extracts at a concentration of 50 mg/ml, incubated at 37°C for 3 hours, and subsequently cryopreserved for 24 hours. Aloe vera extract exhibited the largest inhibition zones against both Gram-positive (30 mm) and Gram-negative (32 mm) bacteria (p < 0.05), followed by mulberry and roselle. After 4 h chilling, the mulberry group showed significantly reduced bacterial counts (p<0.05, 1408 ± 4.8 CFU/mL), whereas post-thaw, aloe vera maintained the lowest bacterial load (p<0.05, 1580 ± 4.9 CFU/mL). Sperm motility and viability were highest in the mulberry group during chilling (p<0.05, 82 ± 3.4% and 85 ± 5.2%, respectively), while aloe vera preserved superior post-thaw motility (p<0.05, 62 ± 5.3%) and viability (p<0.05, 68 ± 7.9%). Roselle extract maintained moderate antibacterial activity and semen quality across both stages. In contrast, the control group without antibacterial additives consistently exhibited the highest bacterial load and poorest sperm parameters. These findings suggest that mulberry extract is a promising alternative to antibiotics in chilled semen extenders, while aloe vera and roselle extracts may serve as effective agents for preserving the quality of cryopreserved goat semen.
